adjuvantの基礎研究

「基礎研究」だからもちろんマウスを使うわけで。免疫反応の惹起はcell-cell contactが必要だったり、cytokineのexcretionが必要だったりするから、in vitroだとやっぱり厳しいのだろう。最近は「○○感染では××というcytokineがあがってくる」とかわかっているから、マウスにある操作を施して、そのあとcytokine levelを測ったりするだけで、どういう系が活性化されたかがわかるのだろう。ただやっぱりT-cell activationをみるのは難しいようで、古典的血清学の手法、血清を薄めていって抗体価をチェックするみたいなのがやりやすいようだ。

10年くらい前の研究には、MF-59がdenderitic cellに取り込まれる過程をimmunofluorecenceを駆使してとらえた実験もあり、こういうところからadjuvant研究は進んできたのか、っと感慨深い。
